چکیده
Despite the great commercial success, Pacific white shrimp Litopenaeus vannamei farming has suffered economic losses due to impact of viral diseases such as White Spot Syndrome. Among innovative approaches mitigate diseases, one most promising is manipulation eukaryote process known interfering RNA (RNAi). This natural mechanism can be induced improve degradation transcripts, through double-stranded molecules (dsRNAs) homologous key target genes. The synthetic production expensive and prevents use this technology on a large scale. In present study, probiotic Bacillus subtilis was engineered produce dsRNAs against WSSV vp28 gene transcripts. results obtained showed that produced by are efficient in inducing RNAi hemocytes. Shrimp pretreated with genetically reprogrammed had higher survival rate when challenged virus, considerable decrease load. Histological analyzes did not identify inclusions survivors' cuticle, gills gastric epithelium. study reports probiotics for antiviral these absorbed at intestine reach shrimp's hemocytes, minimizing replication. current restrictions modified microorganisms, become an important therapeutic tool only aquacultured species, but also treatment human near future.

RNAi and double-strand RNA.
Double-strand RNA (dsRNA) is a signal for gene-specific silencing of expression in a number of organisms. This phenomenon was demonstrated recently in Caenorhabditis elegans when dsRNA was injected into the worm and the corresponding gene products disappeared from both the somatic cells of the organism as well as in its F1 progeny (Fire et al. 1998). This RNA interference, RNAi, has been genera...
متن کاملEnvelope Proteins of White Spot Syndrome Virus (WSSV) Interact with Litopenaeus vannamei Peritrophin-Like Protein (LvPT)
White spot syndrome virus (WSSV) is a major pathogen in shrimp cultures. The interactions between viral proteins and their receptors on the surface of cells in a frontier target tissue are crucial for triggering an infection. In this study, a yeast two-hybrid (Y2H) library was constructed using cDNA obtained from the stomach and gut of Litopenaeus vannamei, to ascertain the role of envelope pro...

متن کاملan introduction to rna interference (rnai)
the discovery of rna interference (rnai) in recent years has revolutionized biological and medical research. rnai is the sequence-specific gene silencing triggered by double-stranded rna (dsrna). until a couple of years ago only biologists working with caenorhabditis elegans and plants could use rnai technology to gain insight into gene function……
